Ready-To-Go PCR Beads Versus GoTaq Green Master Mix: Which PCR Format Fits Your Lab?

Lyophilized PCR bead in a tube next to a liquid master mix

Ready-To-Go PCR Beads vs GoTaq Green Master Mix

Ready-To-Go PCR beads versus GoTaq Green Master Mix is really a choice between two formats: a dry, room-temperature bead and a frozen, ready-to-use liquid. Both let you simply add template and primers, and both deliver the same core reaction chemistry. Where they differ is storage, reaction volume, workflow and what happens after amplification.

This comparison uses each manufacturer's published specifications for Cytiva's illustra™ PuReTaq Ready-To-Go™ PCR Beads and Promega's GoTaq® Green Master Mix. At EVIK, we make lyophilized reagent beads, so we have also included a third option for labs that want bead convenience with their own chemistry.

The Quick Answer

Ready-To-Go PCR Beads

Choose these when room-temperature storage, shipping without a freezer and minimal pipetting matter most, and a fixed 25 µL reaction suits your workflow.

GoTaq Green Master Mix

Choose this when you run routine gel-based PCR, want to load reactions straight onto a gel and need flexibility to change reaction volumes.

Custom Lyophilized Beads

Consider these when you want ambient storage but need your own formulation, qPCR or LAMP chemistry, white-label product or different packaging format.

Format and Storage

This is the biggest practical difference. Ready-To-Go PCR beads are freeze-dried and stored at room temperature in a foil pouch with desiccant, so they need no freezer space and ship without a cold chain. GoTaq Green Master Mix is a liquid that is stored frozen for long-term use and thawed and vortexed before each setup.

For a remote site, a field kit or a lab short on freezer space, that difference can decide the choice on its own. For a busy lab with freezers close to the bench, it matters less.

If you choose beads, read our guide to lyophilized bead handling to learn how to keep them dry and intact. Our article on the features of lyophilized beads covers the wider benefits of the format.

Reaction Setup and Workflow

Ready-To-Go PCR Beads

Each bead is designed for one 25 µL reaction. Add primers, template and water to reach 25 µL, and the bead dissolves into a complete reaction. The pre-dispensed format means fewer pipetting steps and fewer opportunities for error or contamination.

GoTaq Green Master Mix

Because it is a 2X solution, combine equal volumes of master mix and the other reaction components. Promega's protocol gives examples for 25, 50 and 100 µL reactions, so you can scale the volume to suit the experiment.

The trade-off: Beads exchange volume flexibility for speed and consistency, while the liquid mix offers greater flexibility with a few extra pipetting steps.

Reaction Chemistry Compared

The core chemistry is closer than many people expect. At 1X, both systems deliver 1.5 mM MgCl₂ and 200 µM of each dNTP. The differences are in the details: the Ready-To-Go bead uses a pH 9.0 Tris-HCl and KCl buffer with BSA, while GoTaq Green uses a proprietary pH 8.5 buffer.

Both are built around Taq-type polymerases. Promega also offers hot-start and second-generation (G2) GoTaq master mixes as separate products if you need them.

Downstream Analysis

GoTaq Green: Direct-to-Gel Analysis

GoTaq Green Master Mix is designed for direct-to-gel analysis. Reactions are dense enough to load straight onto an agarose gel. The blue dye runs with 3–5 kb fragments in a 1% agarose gel, while the yellow dye runs ahead of the primers (under 50 bp).

If you plan to analyze products by fluorescence or absorbance without purifying them first, Promega recommends its GoTaq Colorless Master Mix, which has the same formulation without the dyes.

Ready-To-Go Beads: Add Your Loading Buffer

Ready-To-Go PCR beads list no tracking dyes, so you add your own loading buffer before running a gel. Cytiva states that the beads are verified for real-time PCR.

Frequently Asked Questions

Is GoTaq Green Master Mix lyophilized?

No. It is a 2X liquid master mix that is stored frozen for long-term use and thawed before setup.

Do Ready-To-Go PCR beads need a freezer?

No. Cytiva specifies storage at ambient room temperature in an airtight foil pouch with desiccant.

Can I load GoTaq Green reactions directly onto a gel?

Yes. The reactions are dense enough for direct loading, and the blue and yellow dyes let you track electrophoresis.

What reaction volume do Ready-To-Go PCR beads use?

Each bead is designed for a 25 µL reaction. GoTaq Green Master Mix can be scaled to different volumes, such as 25, 50 or 100 µL.

Are the reaction chemistries similar?

Yes, at the core. Both give 1.5 mM MgCl₂ and 200 µM of each dNTP in the final reaction, with different buffers: pH 9.0 for the bead and pH 8.5 for GoTaq Green.

Ready-To-Go PCR beads versus GoTaq Green Master Mix: which is better for kits and devices?

For commercial kits and point-of-care devices, a room-temperature bead is often a useful starting point, ideally one formulated and packaged for your own assay. A custom lyophilized bead can be designed around those requirements, subject to formulation feasibility and validation.

Ready-To-Go PCR Beads vs GoTaq Green Master Mix: The Bottom Line

Pick GoTaq Green Master Mix for flexible, gel-based benchwork, and pick Ready-To-Go PCR beads when room-temperature storage and fewer pipetting steps matter more. If neither catalog product fits your assay, volume or brand, a custom lyophilized bead gives you the convenience of the bead format with the chemistry you choose.
